The four biggest moments from ?The Ultimate Fighter?

The second episode of "The Ultimate Fighter" doesn't have the same buzz as the first one, but still had plenty of drama. Before we start, is anyone else thrown off by the extremely close focus on fighters faces? I don't feel the need to count anyone's pores.

On the last show, Luke Barnatt, Team Sonnen's first pick, and Gilbert Smith, from Team Jones, were paired up to fight by Jon Jones. Here are the biggest moments from the show.

Honesty from coaches on Gil's chances to win. They showed a meeting where the coaches spoke about how they were concerned about the fight, and Gil is cognizant of their lack of faith. It gets weirder as his teammates go to the coaches about their concerns on Gil's readiness.

Jones breaking down the overhand punch for Gil. In the past, fighters and coaches hadn't been so explicit about game plans on camera, and didn't give coaching tips like they did on Tuesday night.

A fight that went exactly as his teammates said it would. Gilbert Smith was knocked out by Barnatt, just as his teammates thought. They were worried about his cardio. After he struggled with wrestling against an English opponent in the first round, he was tired in the second. He lazily tried for a takedown, and got Barnatt's flying knee in the face.

"He lost the fight before he got here."
Jones said Smith lost the fight for his lack of confidence, but who can blame him? His coaches and teammates were openly unsure of his ability to fight. He was put up against the top pick on Team Sonnen, but wasn't sure he had anyone behind him.

With Barnatt's win, Team Sonnen got control of the fight and picked Uriah Hall to fight against Team Jones' Adam Cella.

BlackBerry Z10 spotted at UK retailers prior to BB10 event

Looks like retailers might be jumping the gun a bit on the about-to-be-announced BlackBerry Z10 smartphone ahead of the upcoming event. Twitter user @bandozer spotted one at a UK Phones4U retailer and managed to snap a pic (after the break), saying that "it looks and feels good," while our own tipster forwarded us the above image that came from another store. Looks like the only thing that'll stop the numerous leaks is when RIM finally announces the darned thing -- sometime in the next hour or two.

Fed likely to stick to low-rate message this week

WASHINGTON (AP) ? When the Federal Reserve meets this week, it's likely to affirm a message it intends to help lift the economy: that consumers and businesses will be able to borrow cheaply well into the future ? even after unemployment has dropped sharply.

Last month, the Fed signaled for the first time that it will tie its policies to specific economic barometers. It said that as long as the inflation outlook is mild, it could keep short-term rates near zero until unemployment dips below 6.5 percent from the current 7.8 percent.

That could take until the end of 2015, the Fed predicted last month.

The Fed's guidance was designed to give consumers, companies and investors a clearer sense of when super-low borrowing costs might start to rise. Though some key sectors of the economy are improving, analysts think the Fed still feels more time is needed for low rates to spur borrowing, spending and economic growth.

One reason is that many Americans remain anxious about the budget impasse in Washington.

"The Fed is dealing with a lot of uncertainty right now, with all the decisions still to be made on federal budget policy," said Diane Swonk, chief economist at Mesirow Financial, who expects the Fed to make no changes in its support programs when its two-day policy meeting ends Wednesday.

At its December meeting, the Fed said it would keep spending $85 billion a month on bond purchases to keep long-term borrowing costs down. It will continue its bond purchases until the job market improved "substantially."

When it buys bonds, the Fed increases its investment portfolio and pumps more money into the financial system ? something critics say could eventually ignite inflation or create dangerous bubbles in assets like real estate or stocks.

On Friday, when the government will release its jobs report for January, unemployment is expected to remain 7.8 percent. That still-high rate, 3? years after the Great Recession officially ended, helps explain why the Fed has kept its key short-term rate at a record low near zero since December 2008, just after the financial crisis erupted.

In a speech in Ann Arbor, Mich., this month, Chairman Ben Bernanke said he thought too little progress had been made in reducing unemployment and signaled that the Fed's aggressive support programs should continue.

"There is still quite a ways to go," Bernanke said of the unemployment crisis. "There are too many people whose skills and talents are being wasted."

Still, some private economists think the Fed will decide to suspend its bond purchases in the second half of this year. They note that the minutes of the Fed's December meeting revealed a split: Some of the 12 voting members thought the bond purchases would be needed through 2013. Others felt the purchases should be slowed or stopped altogether before year's end.

On one point, economists agree: Once the Fed does decide to scale back its stimulative policies, it will signal its intent well before it actually does so. Policymakers will want to blunt the shocks that could reverberate through financial markets, which have been heavily influenced by the loose-credit policies the Fed has engineered for more than four years.

Interest rates have sunk to record lows. And stock prices have risen as many investors have shifted money into the stock market in search of better returns.

"Nothing will change at this meeting, but as time goes on, I think the Fed will begin laying the groundwork for changes," said Sung Won Sohn, an economics professor at the Martin Smith School of Business at California State University.

Once the Fed does tighten its interest-rate policy, it will inevitably jolt the markets, however much it tries to ease the impact, predicted David Jones, chief economist at DMJ Economic Advisors.

"The second the Fed gives a hint that they are in any way being less accommodative, we will see interest rates shoot higher and stock prices fall," Jones said.

Safer way to vaccinate: Polymer film that gradually releases DNA coding for viral proteins may beat traditional vaccines

Jan. 28, 2013 ? Vaccines usually consist of inactivated viruses that prompt the immune system to remember the invader and launch a strong defense if it later encounters the real thing. However, this approach can be too risky with certain viruses, including HIV.

In recent years, many scientists have been exploring DNA as a potential alternative vaccine. About 20 years ago, DNA coding for viral proteins was found to induce strong immune responses in rodents, but so far, tests in humans have failed to duplicate that success.

In a paper appearing in the Jan. 27 online issue of Nature Materials, MIT researchers describe a new type of vaccine-delivery film that holds promise for improving the effectiveness of DNA vaccines. If such vaccines could be successfully delivered to humans, they could overcome not only the safety risks of using viruses to vaccinate against diseases such as HIV, but they would also be more stable, making it possible to ship and store them at room temperature.

This type of vaccine delivery would also eliminate the need to inject vaccines by syringe, says Darrell Irvine, an MIT professor of biological engineering and materials science and engineering. "You just apply the patch for a few minutes, take it off and it leaves behind these thin polymer films embedded in the skin," he says.

Irvine and Paula Hammond, the David H. Koch Professor in Engineering, are the senior authors of the Nature Materials paper. Both are members of MIT's David H. Koch Institute for Integrative Cancer Research. The lead author of the paper is Peter DeMuth, a graduate student in biological engineering.

Gradual vaccine delivery

Scientists have had some recent success delivering DNA vaccines to human patients using a technique called electroporation. This method requires first injecting the DNA under the skin, then using electrodes to create an electric field that opens small pores in the membranes of cells in the skin, allowing DNA to get inside. However, the process can be painful and give varying results, Irvine says.

"It's showing some promise but it's certainly not ideal and it's not something you could imagine in a global prophylactic vaccine setting, especially in resource-poor countries," he says.

Irvine and Hammond took a different approach to delivering DNA to the skin, creating a patch made of many layers of polymers embedded with the DNA vaccine. These polymer films are implanted under the skin using microneedles that penetrate about half a millimeter into the skin -- deep enough to deliver the DNA to immune cells in the epidermis, but not deep enough to cause pain in the nerve endings of the dermis.

Once under the skin, the films degrade as they come in contact with water, releasing the vaccine over days or weeks. As the film breaks apart, the DNA strands become tangled up with pieces of the polymer, which protect the DNA and help it get inside cells.

The researchers can control how much DNA gets delivered by tuning the number of polymer layers. They can also control the rate of delivery by altering how hydrophobic (water-fearing) the film is. DNA injected on its own is usually broken down very quickly, before the immune system can generate a memory response. When the DNA is released over time, the immune system has more time to interact with it, boosting the vaccine's effectiveness.

The polymer film also includes an adjuvant -- a molecule that helps to boost the immune response. In this case, the adjuvant consists of strands of RNA that resemble viral RNA, which provokes inflammation and recruits immune cells to the area.

The ability to provoke inflammation is one of the key advantages of the new delivery system, says Michele Kutzler, an assistant professor at Drexel University College of Medicine. Other benefits include targeting the wealth of immune cells in the skin, the use of a biodegradable delivery material, and the possibility of pain-free vaccine delivery, she says.

"It's an interesting approach that can be applied not just to delivery of DNA-based vaccine antigens, but other small molecules," says Kutzler, who was not part of the research team.

Eliciting immune responses

In tests with mice, the researchers found that the immune response induced by the DNA-delivering film was as good as or better than that achieved with electroporation.

To test whether the vaccine might provoke a response in primates, the researchers applied a polymer film carrying DNA that codes for proteins from the simian form of HIV to macaque skin samples cultured in the lab. In skin treated with the film, DNA was easily detectable, while DNA injected alone was quickly broken down.

"The hope is that that's an indication that this will translate to large animals and hopefully humans," Irvine says.

The researchers now plan to perform further tests in non-human primates before undertaking possible tests in humans. If successful, the vaccine-delivering patch could potentially be used to deliver vaccines for many different diseases, because the DNA sequence can be easily swapped out depending on the disease being targeted.

"If you're making a protein vaccine, every protein has its little quirks, and there are manufacturing issues that have to be solved to scale it up to humans. If you had a DNA platform, the DNA is going to behave the same no matter what antigen it's encoding," Irvine says.

Jurassic Park 3D Bluray comes home April 23rd after its oneweek theatrical return

We've known since last year that Jurassic Park would make its return to theaters in 3D this April, but now Universal has also dropped the details about its Blu-ray 3D release, as well as the 2D release of all the flicks individually in March. Just to fill in the timeline: first Jurassic Park 1, 2 and 3 will hit Blu-ray/DVD individually (following the triple pack that shipped in 2011) on March 26th, then Jurassic Park in 3D arrives April 5th in regular and IMAX theaters (no, the now-dated CGI has not been redone, you can get a peek in the trailer embedded after the break) followed finally by the Blu-ray 3D April 26th.

While news that the flick is back on the big screen is exciting, we can't help but think there's a few missed opportunities here like the previously mentioned CGI, and lack of any extras detailing the conversion process on the disc. Still, some had complaints about the picture quality of the previous release and it's possible that remastering done in the name of 3D could see a better version this time around for all viewers. Either way, after Titanic, I, Robot and even Top Gun seeing 3D re-release it appears this is definitely a trend. The individual movies are priced at $24.49 on Amazon, and the 3D combo pack is $34.99.

Antigua set to bypass US copyright law with WTO green-lit media ...

In a surprisingly legal move, the government of Antigua is planning to open a website selling media and software without paying any fees to American copyright holders. It comes after the US closed its market to Antiguan gambling companies.

?The small Caribbean nation once thrived on its gambling industry, which at one point employed 5 per cent of its people, TorrentFreak reports. When the US blocked the island's gambling companies from accessing American players, it pulled the rug out from under the whole industry.

Antigua is now set to begin selling music, movies and software without having to pay a cent to the American firms holding copyrights on the wares.

When the US effectively shut down the Antiguan gambling market, the island state sued Washington within the World Trade Organization?? and won. The WTO decided in 2005 that Washington's move violated free trade guidelines, as American companies were allowed free reign domestically.

In 2007, the WTO made a further ruling, allowing Antigua to "suspend" American copyrights for as much as $21 million ever year.

Antigua has informed the WTO of its plan to sell American media online to customers worldwide, even bringing it up at a WTO meeting in December. However, the US struck the topic from discussion, calling it "untimely." The Caribbean state will try again this month, and if all goes according to plan the website is said to be set to go live soon after.

American authorities are hardly signed up to the idea, with the US warning that ?If Antigua actually proceeds with a plan for its government to authorize the theft of intellectual property, it would only serve to hurt Antigua?s own interests" in a December 2012 letter to the WTO.

?Government-authorized piracy would undermine chances for a settlement that would provide real benefits to Antigua. It also would serve as a major impediment to foreign investment in the Antiguan economy, particularly in high-tech industries,? the letter continued.

Mark Mendel, the attorney for Antigua, said the term 'piracy' is irrelevant in this situation, Torrentfreak reports, as the WTO has given Antigua limited immunity to copyright laws.

Facebook Clarifies Developer Policies - Business Insider

Under fire for cutting off popular new apps,?Facebook has announced new rules for developers using its social network.

It is now requiring developers to show "reciprocity," which means their apps must let users easily post content to Facebook, in order to use Facebook's friend-finding features.

The change comes as Facebook abruptly cut off access to Twitter's new Vine video-sharing app and Wonder, a social search app from Yandex, a Russia-based search engine.

For years, Facebook has encouraged app developers on the Web and on mobile devices to use Facebook as a default login and a way of finding friends who are also using the service, a key strategy for making it ubiquitous.

But it has repeatedly clashed with players like Google and Twitter, viewing their attempts to connect with Facebook as an attempt to build up competing social networks at Facebook's expense.

Here's the old rule Facebook had for developers:

Competing social networks: (a) You may not use Facebook Platform to export user data into a competing social network without our permission; (b) Apps on Facebook may not integrate, link to, promote, distribute, or redirect to any app on any other competing social network.

And the new rule:

Reciprocity and Replicating core functionality: (a) Reciprocity: Facebook Platform enables developers to build personalized, social experiences via the Graph API and related APIs. If you use any Facebook APIs to build personalized or social experiences, you must also enable people to easily share their experiences back with people on Facebook. (b) Replicating core functionality: You may not use Facebook Platform to promote, or to export user data to, a product or service that replicates a core Facebook product or service without our permission.

The reciprocity requirement around sharing content and the prohibition on replicating Facebook services are new, but it closely matches a controversial changes Twitter made last year to crack down on apps that mostly acted as clients for reading and posting tweets.

Still, what constitutes "replication," and how easy must sharing be? Facebook, it seems, will decide that.

Vine, Twitter's new video app, allows content to be shared easily, in the form of a link and a preview image. But unlike Facebook-owned Instagram, which injects photos directly into Facebook users' profiles, Vine does not actually post the videos on Facebook.

That mirrors the restricted way with which Instagram interacts with Twitter: Instagram no longer displays photos on Twitter's website or mobile apps, forcing users to click through to the Instagram website to view and interact with them.

Enforcement seems arbitrary. Facebook allows Twitter users to crosspost updates from Twitter to Facebook, but LinkedIn users can no longer crosspost Twitter updates to the professional network. Facebook allows Google's YouTube and Yahoo's Flickr to share photos and videos while using friend-finding features. Twitter likewise allows Flickr users to find Twitter friends, but Instagram users can't.

Consumers pay the price, ultimately, since they must laboriously rebuild their lists of friends on any new service that's innovative enough to threaten dominant players like Twitter or Facebook, or just offered by a company that's not on sufficiently friendly terms with them.

A Facebook spokesperson did not respond to a request to discuss the new policies in greater detail.

Mystery of the Shrunken Proton

Short answer is that I suspect the physics is not new, but something related to something we think we qualitatively know, but we don't really know how to bound the computational errors correctly in a complicated system.

AFAIK, the QED computation techniques that are used to compute bound state of a proton (often modified ordered pertubation methods) aren't particularly convergent so many shortcuts are taken (e.g., use orders of different quantities like non-relativistic velocity, etc). By using a muon and a proton (instead of an electron and a proton), we are essentially replacing something we know more about (the electron) with something we know less about (muon), to try and compute something about something we don't know much about (the proton). Since we don't know much about protons yet, I believe most computations of the bound state are currently just assuming things about them (charge is a point source, nothing about quarks). I haven't read the paper yet, so it's hard to know what they are doing in the QED corrections.

Maybe there is a slight chance that this simplistic system (muon+proton) can macroscopically exhibit something that hints that QCD confinement inside a proton or muon isn't perfect (e.g, the heavy quarks sortof show themselves in a way that we can measure) which would be some interesting new gluon physics that is currently beyond our particle collider reach. But in some ways this might just show us that the QED based adjustments we are making aren't good enough for the real system and we need some even harder to dream up QCD adjustments and it's hard to say that this would definitly be new physics, but perhaps just new math on old QCD physics....

Retrovirus in the human genome is active in pluripotent stem cells

Jan. 23, 2013 ? A retrovirus called HERV-H, which inserted itself into the human genome millions of years ago, may play an important role in pluripotent stem cells, according to a new study published in the journal Retrovirology by scientists at UMass Medical School. Pluripotent stem cells are capable of generating all tissue types, including blood cells, brain cells and heart cells. The discovery, which may help explain how these cells maintain a state of pluripotency and are able to differentiate into many types of cells, could have profound implications for therapies that would use pluripotent stem cells to treat a range of human diseases.

"What we've observed is that a group of endogenous retroviruses called HERV-H is extremely busy in human embryonic stem cells," said Jeremy Luban, MD, the David L. Freelander Memorial Professor in HIV/AIDS Research, professor of molecular medicine and lead author of the study. "In fact, HERV-H is one of the most abundantly expressed genes in pluripotent stem cells and it isn't found in any other cell types."

In the study, Dr. Luban and colleagues describe how RNA from the HERV-H sequence makes up as much as 2 percent of the total RNA found in pluripotent stem cells. The HERV-H sequence is controlled by the same factors that are used to reprogram skin cells into induced pluripotent stem (iPS) cells, a discovery that garnered the 2012 Nobel Prize in Physiology or Medicine. "In other words, HERV-H is a new marker for pluripotency in humans that has the potential to aid in the development of iPS cells and transform current stem cell technology," said Luban.

When a retrovirus infects a cell, it inserts its own genes into the chromosomal DNA of the host cell. As a result, the host cell treats the viral genome as part of its own DNA sequence and begins making the proteins required to assemble new copies of the virus. And because the retrovirus is now part of the host cell's genome, when the cell divides, the virus is inherited by all daughter cells.

In rare cases, it's believed that retroviruses can infect human sperm or egg cells. If this happens, and if the resulting embryo survives, the retrovirus can become a permanent part of the human genome, and be passed down from generation to generation. Scientists estimate that as much as 8 percent of the human genome may be composed of extinct retroviruses left over from infections that occurred millions of years ago. Yet these sequences of fossilized retrovirus were thought to have no discernible functional value.

"The human genome is filled with retrovirus DNA thought to be no more than fossilized junk," said Luban. "Increasingly, there are indications that these sequences might not be junk. They might play a role in gene expression after all."

An expert in HIV and other retroviruses, Luban and his colleagues were seeking to understand if there was a rationale behind where, in the expansive human genome, retroviruses inserted themselves. Knowing where along the chromosomal DNA retroviruses might attack could potentially lead to the development of drugs that protect against infection; better gene therapy treatments; or novel biomarkers that would predict where a retrovirus would insert itself in the genome, said Luban.

Turning these same techniques on the retrovirus sequences already in the human genome, they discovered a sequence, HERV-H, that appeared to be active. "The sequences weren't making proteins because they had been so disrupted over millions of years, but they were making these long, noncoding RNAs," said Luban.

Specifically, the HERV-H sequence was making abundant amounts of RNA in human embryonic stem cells -- and only stem cells. In total, there are more than 1,000 HERV-H retrovirus genomes scattered throughout the human genome. The Luban lab also found high levels of HERV-H RNA in some iPS cells. Other iPS cells, perhaps those lines that were not sufficiently reprogrammed to pluripotency, had lower levels of the HERV-H RNA, another indication that HERV-H may be an important marker for pluripotency.

Interestingly, the HERV-H genes that were expressed in human pluripotent stem cells are only found in the human and chimpanzee genomes, indicating that HERV-H infected a relatively recent ancestor to humans, said Luban.

"Once upon a time HERV-H was an invader to our genome and perhaps caused diseases like AIDS or cancer," said Luban. "Now it seems that a kind of d?tente has been reached. Not only that, but this ancient invader may one day be exploited by clinicians to cure people of a wide range of diseases using stem cell therapies."

Luban and colleagues will next try to determine the specific mechanisms by which HERV-H contributes to pluripotency.

Hepatitis C linked to ink

NEW YORK (Reuters Health) - Researchers are hoping that people will do some research about where to get a tattoo, after a study found a link between body art and hepatitis C.

The new study found that people with the virus were almost four times more likely to report having a tattoo, even when other major risk factors were taken into account, co-author Dr. Fritz Francois of New York University Langone Medical Center told Reuters Health.

Although the study could not prove a direct cause and effect, "Tattooing in and of itself may pose a risk for this disease that can lay dormant for many, many years," Francois said.

About 3.2 million people in the U.S. have hepatitis C, and many don't know because they don't feel ill, according to the Centers for Disease Control and Prevention (CDC).

Hepatitis C is the leading cause of liver cancer and most common reason for liver transplants in the U.S. Some 70 percent of people infected will develop chronic liver disease, and up to 5 percent will die from cirrhosis or liver cancer.

For the current study, researchers asked almost 2,000 people about their tattoos and hepatitis status, among other questions, at outpatient clinics at three New York area hospitals between 2004 and 2006.

Researchers found that 34 percent of people with hepatitis C had a tattoo, compared to 12 percent of people without the infection.

The most common routes of contracting hepatitis C, a blood-borne disease, are through a blood transfusion before 1992 or a history of injected drug use. Injected drug use accounts for 60 percent of new hepatitis cases every year, but 20 percent of cases have no history of injected drug use or other exposure, according to the CDC.

Francois and his colleagues only included people with hepatitis C who did not contract it from these two other common sources.

After accounting for other risk factors, the difference between people with and without hepatitis was even greater, with four times as many tattoos in the infected group than for uninfected people, according to results published in the journal Hepatology.

"This is not a big surprise to me," Dr. John Levey, clinical chief of gastroenterology at the University of Massachusetts Medical School in Worcester, told Reuters Health. Earlier studies had found a link, but they were small and had not taken other risk factors into account as well as this new one did.

"This was one of the stragglers, and now we finally have some numbers for it," said Levey, who was not involved in the study.

Still, the CDC's Dr. Scott Holmberg said the link may not be quite as strong as the findings suggest, because some people who had used illegal drugs probably would not admit it, even on an anonymous questionnaire. And the researchers didn't rule out people who contracted hepatitis before getting their tattoo.

WHAT TO LOOK FOR IN A TATTOO PARLOR

Holmberg, of the CDC's viral hepatitis division, recommends people only have tattoos or piercings done by trained professionals.

"In the U.S., there have been no reports of hepatitis C outbreaks linked to professional tattoo parlors," told Reuters Health by email.

In 2012, 1 in 5 people reported having at least 1 tattoo, according to a Harris poll.

"There are very reputable places that use appropriate standards," said Francois. Tattoo parlors are not federally regulated, and standards vary by state and region, so it's up to the consumer to do their homework, he said.

The Alliance for Professional Tattooists recommend finding a tattoo artist who wears disposable gloves, a clean work space without blood spatters and single-use disposable needle kits.

Levey said he wouldn't prevent his two adult daughters from getting tattoos, but he would make sure they were aware of the hepatitis C risk first.

"A lot of their friends have tattoos, it's the cool thing to do," he said. "They're adults, they can make their own decisions. But I'd mention this to them, because the long-term consequences of hepatitis C are so serious."
